# Apoptosis: Caspase Cascades and the Decision to Die

## Lesson: The cell that dismantles itself

A cell can dismantle itself in an ordered way. Apoptosis packs the cell into neat membrane parcels that neighbors swallow, while uncontrolled death spills the contents and inflames the tissue. One is tidy, the other is messy. Telling them apart means looking at what happens to the contents and the neighbors.

**Example.** The machinery waits in the cell as sleeping procaspases, since building them on demand would be too slow and risky. Initiator caspases sense the damage order and switch on executioner caspases, which cut hundreds of structural targets, and each level multiplies the kill signal. Mitochondria play their part by releasing cytochrome c, which builds the apoptosome that activates the first caspase. Death signals from outside and damage signals from inside converge on the same execution step.

Checkpoints inspect DNA before division, and hand the cell to the death program when damage is beyond repair. Control and self destruction share wiring on purpose. A broken death pathway grows a tumor without any extra division speed, simply by letting cells live that should have died.

**Tip.** Cancer follows when growth, division, and death controls fail together. Escaping programmed death is one of the constraints a tumor must break, and treatment often works by pushing survivors back into apoptosis. Whenever tissue overgrows at a normal division rate, suspect missing deaths before faster births.
