Base editors tether a deaminase to a disabled nuclease and rewrite one letter without cutting both strands. Cytosine editors make C to T changes and adenine editors make A to G changes. Neither cuts both strands, and swaps outside their menu need a different tool. You already know from genome engineering that the cell's repair writes the ending; here repair also decides whether the fresh change sticks or is erased.

Try it together

Suppose your target C sits beside another C inside the editing window. The deaminase acts across several positions near the target, so the neighbour often converts too, giving a bystander edit next to the intended one. Picking a guide that places only the target of the right kind inside the window is the main design skill. Move the window by shifting the guide until the neighbour drops outside it.

Prime editors work differently: they nick one strand and write new sequence from a template carried on the guide itself, which the cell then adopts. They can install all swaps plus small insertions and deletions, and they run cleaner but slower than base editors. A plain nuclease cut is the crudest option, since end joining sprinkles insertions and deletions around the break. Nicking the unedited strand biases repair toward keeping the edit.

Good to know

To choose between tools, match the change to the menu. A C to T change with a clean window calls for a base editor. Any swap, including transversions, or a small insertion calls for prime editing. A plain cut fits knockouts, not precise letters. Compare off-target habits too: base editors risk stray deamination, prime editors run more precise, and plain cuts add indel errors from repair.
