How a Chromatographic Separation Works

A mixture separates because each component divides itself differently between a moving phase and a stationary one. Retention time is the record of that division, and the plate model turns it into something that can be predicted.

Every chromatography setup has the same two actors: a still stationary phase and a flowing mobile phase. Each component of the mixture splits its time between the two. Strong stickers linger on the still phase while easy riders race ahead with the flow, so different balances give different speeds and the mixture stretches into separate bands.

Retention time is the record of that split. An unretained component washes straight through, and its arrival marks the dead time every other time is measured against. The retention factor k equals the extra wait divided by the dead time: with a dead time of 1 minute and a peak at 4 minutes, k equals 3. A bigger k means the component spent more time stuck to the stationary phase.

Try it together

You can watch this on paper. Put the sample dot above the solvent level so the pool never washes it away, let capillary action climb the strip, then dry it and read the bands. How far each band travelled records exactly how that component divided itself between moving and staying put under that pair of phases.

Good to know

The plate model turns the column into a stack of tiny equilibration stages: more plates mean sharper peaks. Polarity is your steering wheel: a polar stationary phase grips polar components longer, so swapping it for a nonpolar one shortens their stay and reshuffles the order.
