Absorbance is proportional to concentration and path length, which makes a spectrophotometer a concentration meter. The proportionality fails in predictable ways, and knowing where it fails is what keeps the numbers honest.

Absorbance is proportional to concentration and path length, which makes a spectrophotometer a concentration meter. The law reads A equals epsilon times l times c, where epsilon scores how eagerly the chemical soaks up that colour. Once you know epsilon, flip the equation to read any unknown concentration straight off its absorbance. A solution of 0.01 moles per liter in a 1 cm tube with absorbance 2 gives epsilon 200.

Choose the peak of the spectrum for analysis. There the chemical drinks in that wavelength most eagerly, so even tiny amounts move the reading. A high epsilon means a sensitive test, while a low one needs plenty of chemical before absorbance stirs. Match the wavelength to the peak and sensitivity follows.

Try it together

Transmittance and absorbance tell one story on two scales. Transmittance is the fraction of light getting through, and absorbance is minus log of that fraction. Full light through means absorbance 0, a tenth through means 1, and a hundredth means 2. Each absorbance unit passes ten times less light, which is the logarithmic link in action.

Good to know

Respect where the law fails. At high concentration the straight line bends, and stray light reaching the detector bends it further. A sample above the top standard cannot be trusted, so dilute it into range and remeasure. Never extend the line beyond tested standards.

Absorbance scales with amount, logs link it to light, and the line only rules its tested range.
